摘要
从孔雀绿污染土壤中分离筛选出1株对孔雀绿染料有强脱色能力的菌株MGT3,根据其形态学特征和生理生化鉴定以及16S rDNA序列分析,初步鉴定为无色杆菌属(Achromobacter sp.)的菌株.在振荡培养条件下对该菌株的脱色反应条件进行了研究,结果表明,MGT3菌株的最适脱色pH为7,最适脱色温度为30℃,孔雀绿初始浓度小于50mgL-1时有最佳脱色率.在最适脱色条件下脱色8h,孔雀绿脱色率可达到90%以上.利用薄层层析法、紫外-可见分光光度计法、高效液相色谱法对孔雀绿脱色代谢产物进行了分析,结果表明,孔雀绿在脱色过程中被降解,生成了其他代谢产物.
A strain, with strong decoloring capability for malachite green, was isolated from the malachite green contaminated soil and named as strain MGT3. It was identified as Achromobacter sp. by its morphological, physiological and biochemical characters and 16S rDNA sequence analysis. The results from the study on the decoloring conditions of strain MGT3 showed that the optimum temperature for decolorization was 30℃, and optimum pH was 7 under shaking decolorization situation. The best decolorization rate could be obtained at the initial concentration with malachite green less than 50 mg L--1 . Under the optimum conditions, the strain could remove 90.6% of the color of malachite green in 8 h treatment. The results of TLC, UV- VIS and HPLC showed that most of malachite green could be degraded by strain MGT3. Fig 8, Tab 1, Ref26
