Establishment of a Model of Transplanting Kidney lschemia-reperfusion Injury in Rats

摘要 【目的】通过大鼠自体原位肾移植建立移植肾缺血-再灌注损伤(IRI)动物模型,为进一步探讨移植肾IRI的发病机制及防治措施奠定基础。【方法】雄性SD大鼠36只随机分成对照组(n=6)、手术组(n=18)和假手术组(H-12)。观察手术组和假手术组大鼠术后1,3,5,7d[血尿素氮(BUN)、血肌酐(Cr)]的变化并与对照组比较,观察术后24h肾脏的形态学变化。【结果】对照组BUN平均(7.1±0.6)mmol/L,Cr(60.3±3.1)μmol/L;手术组术后1、3、5dBUN平均分别为(38.9±6.9),(31.8±6.6)和(16.8±5.3)mmol/L,Cr平均分别为(287.3±26.6),(201.0±26.3)和(132.2±24.7)μmol/L,与对照组相比差异均有显著性意义(F〈0.05),至d7肾功能恢复正常,手术成功率88.9%;假手术组术后肾功能变化较小,与正常值相比差异无显著性意义(P〉0.05);术后24h形态学观察,手术组大鼠肾脏呈典型的IRI改变,假手术组与对照组犬鼠肾脏未见明显病理改变。【结论】该大鼠移植肾IRI模型的建立操作简单、手术难度低、稳定性良好、成功率高,是研究移植肾IRI的理想模型。 [Objective] To establish a model of transplanting kidney ischemia-reperfusion injury(IRI) in rats with auto orthotopic kidney transplantation, and to make a preparation for further investigation on the mechanism of transplanting kidney IRI and the prevention and treatment of the injury. [Methods] Male Sprague-Dawley rats( n =36) were randomly divided into 3 group: control group( n =6) ,operation group( n = 18) and sham operation group( n = 12). The renal function of 1,3,5, 7 days after operation of operation group and sham operation group were observed and compared with that of the control group. The pathological changes 24h after operation were also observed. [Results] The mean values of blood urea nitrogen(BUN) and serum creatinine(SCr) were (7.1±0.6)mmol/L and (60.3±3.1)μmol/L in the control group, respectively. In the operation group 1,3,5 day after operation., the mean values of BUN were (38.9±6.9) ,(31.8±6.6) and (16.8 ±5.3)mmol/L, respectively, and the mean values of SCr were (287.3±26.6), (201.0±26.3) and (132.2±24.7)μmol/L, respectively. They were all significantly different from those in the control group ( P〈0.05). Till the seventh day, the renal function became normal. The successful rate of operation was very high (88.9%). The change of renal function was slight in the sham operation group, as compared with normal value( P〉0.05). For the morphological examination, typical pathological changes of IRI were observed in the operation group, while those of the sham operation group and the control group were unobvious. [Conclusion] The model of transplanting kidney IRI has advantages of mild operative difficulty, better stability, reliability and with high successful rate,so it is an ideal model for IRI studies in kidney transplantation.
