野生榛蘑多糖提取、分离纯化和相对分子量的测定 被引量:2

Extraction,isolation and purification of polysaccharides in Armillaria mellea and determination of its molecular weight

摘要 野生榛蘑多糖采用水提法得到多糖粗品,经Sevage法除蛋白,H_2O_2法脱色素和透析法去除小分子后,采用DEAE-纤维素柱层析纯化,分离得三种多糖成分Ⅰ,Ⅱ和Ⅲ.纯化得的榛蘑多糖经葡聚糖凝胶层析法测得相对分子量分别为132429,93917,74687 Da. Crude polysaccharides of Armillaria mellea extracted with water were purified by Sevage method to remove protein, H2O2 method to remove pigment and dialysis remove small molecules. The DEAE cellulose column chromatography was continue to be used for polysaccharides purification. The results showed that Armillaria mellea polysaccharide is composed of Ⅰ, Ⅱ and Ⅲ. Its molecular weight was determined by sepharose gel filtration and the molecular weight of Armillaria meUea polysaccharide was 132429 Da, 93917 Da and 74687 Da.
