根癌农杆菌介导的章丘大葱遗传转化体系研究 被引量:3

摘要 建立了根癌农杆菌介导转化章丘大葱愈伤组织的体系,即用OD600值为0.6的农杆菌悬浮液浸泡大葱愈伤组织,侵染30 min,共培养3 d时的转化效果最佳。获得的愈伤组织GUS基因瞬时表达率为33.92%,PCR检测结果表明,转化率为0.59%。 Based on the Agrobacterium -mediated T- DNA insertion, an efficient transformation system of Alliumfistulosum L was established. The highest transformation rate was obtained under the conditions of the Agrobacterium tumefaciens' OD600= 0. 6, infection for 30 minutes and co - culture for 3 days. The transient expression rate of GUS gene was up to 33.92% over all. The PCR analysis showed that the target gene was integrated into the genome of Allium fistulosum L. with the conversion rate of 0.59%.
