摘要
目的:探讨血小板源性生长因子(PDGFs)及胰岛素样生长因子(IGFs)对大鼠椎间盘退变(IDD)的影响。方法:36只成年健康SD大鼠,随机分为PDGFs实验组、PDGFs加IGFs实验组及阴性对照组3组,通过手术方法建立IDD模型。在术后不同时期对3组分别注射PDGFs、PDGFs加IGFs及生理盐水,于造模后1个月和2个月每组分别处死6只大鼠,完整取出L3-4椎间盘测量蛋白多糖含量,取L4-5椎间盘使用免疫组化法检测Ⅱ型胶原含量,取L5-6椎间盘使用流式细胞仪检测各组大鼠椎间盘细胞的凋亡情况。结果:造模当日和术后1个月开始注射试剂的3组椎间盘蛋白多糖含量、Ⅱ型胶原含量及细胞凋亡率差异均有统计学意义(均P〈0.05)。结论:PDGFs对大鼠IDD有预防和治疗作用,PDGFs与IGFs联用有协同作用。
Objective To study the effect of the platelet-derived growth factors(PDGFs) and the insulin-like growth factors(IGFs) on intervertebral disc degeneration in the rat's model by operation.Methods 36 adult healthy rats were randomly divided into three groups: experimental group of PDGFs,experimental group of PDGFs with IGFs and negative-control group.Each group has 12 rats.Degenerative rat model of three groups was established by destructing their lumber spines.The experimental group rats were randomly divided into two groups which were intraperitoneal injected with PDGFs(100 ng·kg^-1) or PDGFs(100 ng·kg^-1) and IGFs(100 ng·kg^-1) on the day of operation or on the day one months post-operation respectively.The injection repeated every the other day until they were killed,and in the negative-control group 0.9% NS of the same dosage of PDGFs were injected.Six rats were killed from each group one or two month after the operation respectively.The tissue of L3~4,L4~5,L5~L6 discs en bloc were acquired.The PG content of the L3~4 disc was measured by phloroglucin,the Ⅱcollagen content of the L4~5 disc was measured by immunohistochemistry respectively and the apoptosis cells of the L5~6 disc were assayed by flowcytometry.Results One month after operation,the proteoglycan contents,the Ⅱcollagen content and the apoptosis cells rates had significant difference among the experimental group of PDGFs,the experimental group of PDGFs with IGFs and negative-control group(P〈0.05).Two months after operation,the content of proteoglycan,the Ⅱcollagen content and the apoptosis cells rates had significant difference among experimental group of PDGFs,the experimental group of PDGFs with IGFs and negative-control group(P〈0.05).Conclusion PDGFs,synergetic with IGFs,prevents the intervertebral discs from degeneration of the rat's model.
