摘要
目的研究当归和川芎对尾吊大鼠比目鱼肌(SOL)肌球蛋白重链(MHC)表达及肌萎缩的影响。方法尾部悬吊法建立模拟失重模型,将大鼠分为同步饲养组(Con)、尾吊灌溶媒组(HLU+W)、尾吊当归灌胃组(HLU+Ang)和尾吊川芎灌胃组(HLU+Lig)。免疫组化法检测大鼠比目鱼肌肌球蛋白重链的表达。结果与同步饲养组相比,尾吊组比目鱼肌的湿重,肌重体重比以及I型和II型肌纤维横截面积均有所下降,而MHCII的表达和II型肌纤维的比例上升。与未治疗组相比,当归可以使I型和II型肌纤维横截面积分别提高29%和35%,MHCII的表达下降。川芎可使I型肌纤维横截面积提高25%(P=0.064)并相应减少MHCII的表达。结论当归和川芎均能显著抑制模拟失重导致的MHCII型表达的升高,并缓解尾吊大鼠肌纤维横截面积的缩小。
Objective To study the effects of two Traditional Chinese Herbs (Angelica sinensis and Ligusticum) as a countermeasure on muscle atrophy and myosin II heavy chain (MHC II) expression in soleus fibers of arat model (tail-suspended) hind-limb unloaded. Methods Six rats each were assigned to a 4-group design:control group (Con),other 3 groups respectively to be hind-limb unloading plus intragastric instillation of water(HLU+W),of Angelica sinensis decoction (HLU+Ang) and of Ligusticum decoction (HLU+Lig). The myosin II heavy chain (MHC II) expression in soleus fibers were measured with immuno-histo-chemical method. Results As all 3 hind-limb unloading groups ( HLU+W,HLU+Ang and HLU+Lig) compared to Con,in soleus muscle wet weight (MWW),muscle wet weight to body weight ratio (MWW/BW) and type I and II fiber CSA decreased,whereas MHC II expression and the percentage of type II fibers increased. Compared to untreated group(HLU+W),Angelica sinensis enhanced type I and II fiber CSA by 29% and 35% respectively,expression of MHC II (type II fiber percentage) decreased as well as Ligusticum increased type I fiber CSA by 25% (P=0.064) and reduced MHC II expression (decreased type II fiber percentage). Conclusion Both Angelica sinensis and Ligusticum can alleviate the weightlessness-induced shift from slow-twitch to fast-twitch fibers. Angelica sinensis also can alleviate CSA reduction of soleus muscle in hind-limb unloaded rats.
