摘要 目的检测Notch-1、Jagged-1及p-mTOR三种蛋白在胰腺癌组织中的表达并探讨它们与临床病理特征的关系及三者之间的相关性。方法采用免疫组织化学SP法检测40例胰腺癌组织和20例正常胰腺组织中Notch-1、Jagged-1及p-mTOR三种蛋白的表达。结果Notch-1的阳性表达主要位于肿瘤细胞胞膜和胞浆,Jagged-1、p-mTOR的阳性表达主要位于细胞浆。Notch-1、Jagged-1及p-mTOR在胰腺癌组织中的阳性表达率分别为80%、75%和72.5%,均显著高于正常胰腺组织中的表达,差异有统计学意义(P<0.05)。Notch-1、Jagged-1及p-mTOR的异常表达均与胰腺癌的淋巴结转移和TNM分期有关(P<0.05),与患者的性别、年龄、部位、分化程度和病理分型无关(P>0.05)。Notch-1、Jagged-1及p-mTOR三者间的表达均呈正相关。结论Notch-1、Jagged-1及p-mTOR的过表达促进胰腺癌的发生发展、浸润转移,Notch-1信号通路可能间接调节mTOR的活性。 Objective To detect the expressions of Noteh-1, Jagged-1 and p-mTOR protein in pancreatic carcinoma and to determine their clinieopathological significance and the correlation among the three protein expressions. Methods SP immunohistoehemical method was used to detect the expression of Noteh-1, Jagged-1 and p-mTOR in twenty eases of normal pancreas and forty cases of pancreatic carcinoma tissues. Results The positive staining for Notch-1 was shown yellow and predominantly observed in membrane and cytoplasm of tumor cells. The positive staining of Jagged-1 and p-mTOR was shown yellow in cytoplasm. The positive expression rates of Notch-1, Jagged-1 and p-roTOR were 80% , 75% and 72.5% respectively in pancreatic carcinoma tissues, which were notablely higher than that in normal pancreatic tissues. The aberrant expressions of Notch-1, Jagged-1 and p-mTOR were associated with lymph node metastasis and TNM stages but not with the gender, age, location, differentiation degree and pathologic type. A highly posi- tive correlation was found among the three protein expression. Conclusion The up-expression of Notch-1, Jagged-1 and p-mTOR can promote the growth, development and metastasis of pancreatic carcinoma. Notch-1 signaling pathway may regulate the activation of mTOR indirectly.
