MMP-9及NF-κB对人颅内动脉粥样硬化斑块稳定性的影响 被引量:14

摘要 目的探讨MMP-9、核因子κB(nuclear factor kappa B,NF-κB)在人颅内动脉粥样硬化斑块中的表达特点及其与粥样斑块稳定性的关系。方法收集脑梗死死亡患者的颅内Wills动脉环标本19例,常规石蜡包埋、切片、HE染色,根据镜下结构将斑块组织分为不稳定性斑块组和稳定性斑块组;采用免疫组织化学方法观察MMP-9、NF-κB在动脉粥样硬化斑块中的表达情况。结果MMP-9、NF-κB在不稳定性斑块中的表达较在稳定性斑块中的表达均明显增加(P<0.01);二者的表达呈明显的正相关(r=0.763)。结论MMP-9、NF-κB是促进动脉粥样硬化斑块不稳定的重要因素,NF-κB可能促进了MMP-9的表达。 Objecive To observe the expression of matrixmetalloproteinases-9 (MMP-9) and NF-κB in intracranial atherosclerotic plaques in human and investigate the relationship between MMP-9 and NF-κB and plaque stability. Methods 19 specimen of human Willis arterial circle atherosclerosis plaques were obtained from patients undergoing fatal cerebral infarction, and then were routine paraffin embedded, sectioned at 2 μm , and stained with hematoxylin and eosin. They were divided into stable and unstable plaques according to the structure under the optical microscope, the quantities of MMP-9 and NF-κB were performed by using immunohistochemical staining technique. Results The protein expression of MMP-9 and NF-κB in the unstable plaques was much stronger than that in the stable ones. A positive correlation was observed between the expression of MMP-9 and the NF-κB. Conclusion MMP-9 and NF-κB are important factors in the promotion of unstable plaque and NF-κB may promote the expression of MMP-9.
