ADAM33基因T1位点多态性与支气管哮喘发病的相关性 被引量:12

Association of Polymorphism of T1 Locus Allele in ADAM33 Gene with Bronchial Asthma

摘要 目的探讨广州市儿童ADAM33基因T1位点多态性与儿童支气管哮喘发病的相关性。方法从广州市海珠区、白云区和花都区共8所小学1~6年级7635例学生中筛选获得92例哮喘儿童(哮喘组)及91名健康儿童(健康对照组),采集2组儿童空腹静脉血2mL,应用试剂盒提取其基因组DNA,进行聚合酶链反应,反应产物进行限制性片段长度多态性(PCR-RFLP)分析,并对部分扩增样品测序。采用SPSS13.0软件进行统计学分析。结果ADAM33基因T1位点Met764/Met764、Met764/Thr764、Thr764/Thr7643种基因型在哮喘组分布频率分别为77.2%(71/92例)、20.7%(19/92例)、2.2%(2/92例),健康对照组分布频率分别为87.9%(80/91例)、11.0%(10/91例)、1.1%(1/91例);哮喘组各基因型分布频率与健康对照组比较差异无统计学意义(χ2=3.66P>0.05)。ADAM33基因T1位点Thr764等位基因在哮喘组与健康对照组分布频率分别为12.5%、6.6%,哮喘组与健康对照组ADAM33基因T1位点等位基因频率比较差异无统计学意义(χ2=3.69P>0.05)。结论ADAM33基因T1位点基因多态性对广州市儿童哮喘发病的影响不明显。 Objective To explore the association of the polymorphism of T1 locus allele in ADAM33 gene with bronchial asthma in children in Guangzhou. Methods From 8 primary schools ( Grade 1 - Grade 6 school children) in Guangzhou three urban districts ( Haizhu, Baiyun and Huadu Districts) ,92 screened patients with asthma (asthma group) and 91 unrelated healthy children were selected (healthy control group) for fasting blood,genome DNA by kit ;polymerase chain reactionrestriction fragment length polymorphism (PCR-RFLP) and DNA sequencing was used to determine polymorphism of T1 locus allele in ADAM33 gene of children in both groups. SPSS 13.0 software was used to analyze the data. Results The frequencies of the genotypes( Met764/Met764, Met764/Thr764 ,Thr764/Thr764) were 77.2% (71/ 92 ) ,20.7% (19/92) ,2.2% (2/92) , respectively, in asthma group, and 87.9% (80/91) , 11.0% (10/91) , 1.1% (1/91) , respectively, in healthy control group. No significant difference was found in the distributions of the genotype ( Met764/Met764, Met764/Thr764, Thr764/ Thr764) between asthma group and healthy control group (χ^2 = 3.66 P 〉 0.05 ). The frequencies of alleles (Thr764) were respectively 12.5% in asthma group and 6.6% in control healthy group. No significant difference was found in the allele (Met764,Thr764) frequencies between the bronchial asthma group and healthy control group(χ^2 = 3.69 P 〉0. 05 ). Conclusion The result suggests that T1 locus genetic polymorphism is weakly associated with asthma.
