摘要
目的通过检测子痫前期患者β-纤维蛋白原-455基因,分析其基因多态性及子痫前期发生的关系。方法利用聚合酶链反应,限制型长度多态性对2007年1至12月在广东省妇幼保健院分娩的96例子痫前期患者和98例正常孕妇进行不同β-纤维蛋白原-455G/A基因型分析,并观察两组血浆纤维蛋白原情况。结果子痫前期患者血浆纤维蛋白原水平显著高于正常妊娠者(P〈0.05)。β—FIB-455G/A基因多态性GA+AA等位基因携带者血浆纤维蛋白原水平明显高于GG等位基因者(P〈0.05),可见A等位基因与高纤维蛋白原水平相关。A等位基因频率及GA、AA基因型在子痫前期组和对照组间无明显区别(P〉0.05)。结论β—FIB-455G/A基因多态性A等位基因可能不是子痫前期的遗传易感基因,但与子痫前期患者血浆高纤维蛋白原水平相关。
Objective To evaluate the polymorphism of different alleles in β-455G/A gene, the distributive characteristics in pre-eclampsia patients and normal pregnant women, and the relationship of these genetic variants with plasma fibrinogen levels. Methods 96 pre-eclampsia women, and 98 normal pregnant women , their plasma levels offibrinogen were determined. Polymorphisms of β -455G/A gene were measured by polymerase chain reaction(PCR) and restriction fragment length polymorphism(RFLP) and HhaⅢ endonuclease digestion and Polyacry- lamide Gel Electrophoresis(PAGE).Results Comparing the plasma fibrinogen level, a significantly plasma fibrinogen concentration was founded in re-eclampsia group(P 〈 0.05). Comparing the plasma fibrinogen level between the different genotype group, a significantly higher plasma fibrinogen level in the β-FIB-455G/AGA+AA group was detected(P〈 0.05). There was no significant difference in the distribution of the Bβ-FIB-455G/A gene between two groups. Conclusions Allele distribution of Bβ-FIB-455G/A was significantly associated with plasma fibrinogen level.The variation frequency offihrinogen β-455G/A genotype was not associated with the occurrence of pre-eclampsia.
