摘要
目的观察柴芩肾安方对IgA肾病(IgAN)大鼠肝细胞生长因子(HGF)表达的影响。方法通过灌服并定时静脉注射牛血清白蛋白(BSA)复合感染葡萄球菌肠毒素B(SEB)的方法制成IgAN模型,用柴芩肾安方治疗,以氯沙坦为对照。第15周末,检测大鼠24 h尿蛋白量(Upr)、血清肌酐(Scr)和尿素氮(BUN);观察大鼠肾组织病理改变情况;采用免疫组化和RT-PCR法分别检测肾组织HGF蛋白及其mRNA的表达。结果模型组大鼠肾小球系膜增生,伴见弥漫的IgA和少量IgG沉积;Upr、BUN和Scr较正常组明显升高(P<0.01)。两治疗组大鼠肾小球系膜增生和IgA、IgG的沉积减轻;Upr、Scr和肾组织HGF蛋白与模型组比较,有显著性差异(P<0.01,P<0.05),且柴芩肾安方组BUN和肾组织HGF mRNA与模型组比较,有显著性差异(P<0.01)。结论柴芩肾安方可能通过促进肾组织HGF蛋白及其mRNA的表达调节细胞外基质(ECM)代谢,从而延缓IgAN大鼠肾脏病变进展。
Objective To observe the impacts of Chai Qin Shen An Fang(CQS) on expression of hepatocyte growth factor(HGF) in IgA nephropathy(IgAN) rats. Methods IgAN rat models were established by administrating bovine serum albumin(BSA) and injecting staphylococcus enterotoxin B (SEB)into phallic vein. Two treatment groups were prepared with CQS and losartan( Los )involved as control. At the end of the 15th week,the levels of 24- hour urinary protein(Upr), serum creatinine (Scr)and blood urine nitrogen (BUN) were measured. Pathologic changes of renal tissues were observed. The pathological change of renal tissue of rats was observed. Immunohistochemistry and RT - PCR were used to examine respectively the pro- tein and mRNA expressions of HGF in renal tissues. Results The mesangial cells and matrix increased and also diffused IgA with little IgG depositions in glomerulus were found in model group. Upr, BUN and Scr in model group were higher than those in normal group( P 〈 0.01 ). The mesangial proliferation and deposits of IgA and IgG were attenuated in two treatment groups. The significant difference was presented in Upr, Scr and HGF protein in kidney tissue in comparison of model group with two treatment groups( P 〈 0.01 ,P 〈 0.05 ). Comparing with model group, the levels of BUN and HGF mRNA in kidney tissue in CQS group were different significantly(P 〈 0.01 ). Conclusion CQS can improve the protein and mRNA expressions of HGF so as to retard the development of renal pathologic changes in IgAN rats.
