摘要
采用Plackett-Burman试验设计法及响应面法分析,对nsin高产菌株Hs-5进行了发酵工艺条件的优化研究。利用Plackett Burman试验设计筛选出影响产nisin效价的3个主要因素,即接种量、培养温度和蔗糖添加量。在此基础上用最陡爬坡路径逼近最大响应区域。结果表明,接种量和这趟添加与Nisin效价存在极显著的相关性,通过求解回归方程得到优化主要发酵条件:接种量5%,培养温度31℃,蔗糖添加量25 g/L。经培养验证,预测值与验证实验平均值接近,在此优化条件下Nisin效价可达到2859。
SAS8.0 the help of software, using Plackett-Burman test design and respome surface analysis, rain high-yield strains of Hs-5 for the fermentation process to opfmize the conditions for research.
