摘要 采用Plackett-Burman试验设计法及响应面法分析,对nsin高产菌株Hs-5进行了发酵工艺条件的优化研究。利用Plackett Burman试验设计筛选出影响产nisin效价的3个主要因素,即接种量、培养温度和蔗糖添加量。在此基础上用最陡爬坡路径逼近最大响应区域。结果表明,接种量和这趟添加与Nisin效价存在极显著的相关性,通过求解回归方程得到优化主要发酵条件:接种量5%,培养温度31℃,蔗糖添加量25 g/L。经培养验证,预测值与验证实验平均值接近,在此优化条件下Nisin效价可达到2859。 SAS8.0 the help of software, using Plackett-Burman test design and respome surface analysis, rain high-yield strains of Hs-5 for the fermentation process to opfmize the conditions for research. First of all, the use of Phckett Burman experimental design selected affect production of titres nisin three main factors, namely, inoculation, temperature and amount of sugar added. On this basis, with the steepest climbing path in response to the largest regional approach, using Box-Behnken experimental design and response surface analysis regression analysis. The results showed that the amount of coverage and add the purpose of his rosin and potency of the existence of significant relevance, by solving the regression equation optimized fermentation of the main conditions: inoculum of 5%, temperature 31℃, add the amount of sucrose 25 g/L. After training verification, validation and test predictive value dose to the average in the optimum conditions nisin potency can be reached 2859.
