大豆异黄酮对代谢综合征模型大鼠血脂及肝脏PPARα基因的影响 被引量:9

Effects of soybean isoflavones on expression of PPARα mRNA and blood lipid level in metabolic syndrome rats

摘要 目的研究大豆异黄酮对代谢综合征(MS)模型大鼠血脂及肝组织过氧化物酶体增殖物激活受体α(PPARα)基因表达的影响。方法将60只雄性SD大鼠随机分为普通饲料组(10只)和高脂高盐高糖饲料组(50只),20周后成功复制MS大鼠模型37只,将37只MS模型大鼠随机分为模型组(9只),大豆异黄酮(SI)高、低剂量组(各10只)以及非诺贝特组(8只),灌药4周后,用生化法测血脂,实时荧光定量PCR方法测定各组大鼠肝脏PPARαmRNA的表达情况。结果与模型组相比,SI高剂量组血清TG、TC以及LDL-C水平明显降低,并且肝脏PPARαmRNA的表达增加,与非诺贝特组相比未见显著差异。结论大豆异黄酮对MS模型大鼠脂代谢紊乱具有调节作用,其机制可能与激活PPARαmRNA的表达有关。 [Objective] To study the therapeutic effects of Soybean Isoflavones on the expression of peroxisome proliferator activated receptor α (PPARα) and blood lipid level in metabolic syndrome rats. [Methods] Sixty male SD rats were randomly divided into normal diet group (NC, n =10) and high-fat diet group (n =50) to induce metabolic syndrome model. Rats were fed with high high-lipid high-salt high-sugar food for 20 weeks except NC group rats. After 20 weeks we got 37 metabolic syndrome rats successfully, and then these 37 metabolic syndrome rats were randomly divided into MS model group (MS, n =9), Fenofibrate-treated group (FG, n =8) and two SI-treated groups (SIH, SIL, n =10). SI 360, 90 mg/kg were given in two SI treatment groups. Blood lipid level was measured after 4 weeks and mRNA level of PPARα of rat hver was determined using FQ-PCR (Fluorescent Quantitative PCR). [Results] In high-dose SI treatment group, both the LDL-C and TC levels were significantly lower than those in MS group (P 〈0.01), the TG was lower too (P 〈0.05), while the expression of PPARα was increased compared with HC group. [Conclusion] SI can regulate the blood hpid level in metabolic syndrome rat, and the mechanism may be that SI can up-regulate the expression of PPARα.
