摘要
以麻疯树籽饼为原料,研究了麻疯树籽饼中粗蛋白的提取工艺,并在此基础上采用超滤技术对毒蛋白Curcin进行脱除。试验结果表明:在提取剂为含1.00 mol/L NaCl的0.005 mol/L PBS缓冲液、原料粒度80目、提取时间90 min、料液比1∶12、提取温度65℃下粗蛋白提取率为98.11%;在超滤时间60 min、超滤压力0.14 MPa、超滤温度45℃、超滤液质量浓度3.235 mg/mL的条件下,毒蛋白Curcin脱除率达到97.62%。
The extraction technology of crude protein from the cake of Jatropha curcas L. seed and the removal of toxic protein Curcin by ultrafihration technique were studied. The results showed that the optimal extraction conditions were as follows :0.005 moL/L PBS buffer containing 1.00 mmol/L sodium chloride used as extraction solvent, particle size of the material 80 meshes, extraction time 90 min, ratio of solid to liquid 1:12 ,extraction temperature 65 %. Under the optimal conditions ,the extraction efficiency of crude protein reached 98.11%. The removal rate of the toxic protein Curcin was 97.62% under the conditions of ultrafihration time 60 min, temperature 45 ℃, uhrafiltration pressure 0.14 MPa and the mass concentration of uhrafiltrate 3. 232 mg/mL.
