摘要
以大孔微球硅胶(30nm,8μm)作基体,经γ-氨丙基三乙氧基硅烷活化与间隔臂氯代 环氧丙烷反应后,再与预先合成的四(对羟基苯基)卟啉配体键合,构成电荷转移型亲和色谱 固定相。以pH=7.0的磷酸盐缓冲溶液作流动相,成功实现对一些核苷酸的高效液相亲和色 谱分离。
5,10,15,20-Tetrakis (4-hydroxyphenyl) porphine (THPP ) had been synthesized and used as a stationary phase ligand. A wide-pore(30 nm), microspheric silica(8μm) as the matrix activated by γ aminopropyltriethoxysilane, then reacted with spacer arm-epichlorohydrin, and bonded with the THPP li- gand to produce a new stationary phase of charge-transfer type-THPP-silica. When phosphate buffer(pH =7. 0) was used as the mobile phase, the separation of nucleosides and nucleotides was achieved by the detection of ultraviolet detector (254 nm).
