应用RNA干扰技术研究AKT和ERK2基因对子宫内膜癌细胞自噬活性的影响 被引量:1

摘要 自噬是真核细胞蛋白质降解的主要途径,参与调控细胞的生长。研究发现,自噬活性的变化与恶性肿瘤的发生、发展有关,子宫内膜癌细胞的自噬活性明显减弱。另有报道,在子宫内膜癌中存在AKT和ERK2基因的高表达。迄今,有关AKT和ERK2基因对子宫内膜癌细胞自噬活性是否具有调节作用仍然不清楚。本研究应用RNA干扰(RNAinterference,RNAi)技术分别沉默子宫内膜癌细胞AKT和ERK2基因的表达,观察AKT和ERK2基因表达下调后对子宫内膜癌细胞自噬活性(以LC3蛋白的表达量表示)的影响,以阐明子宫内膜癌发生、发展过程中自噬活性的改变及其调节机制,为通过于预相关信号蛋白以调节自噬活性进而治疗子宫内膜癌奠定理论基础。
