摘要
目的:探讨表皮生长因子受体(Epidermal growth factor receptor,EGFR)mRNA在膀胱移行细胞癌(transitional cell carcinoma,TCC)患者外周血中的表达及意义。方法:收集62例膀胱移行细胞癌患者外周静脉血及癌组织标本,提取总RNA,行RT-PCR,检测EGFR mRNA的表达。25例健康人外周静脉血标本作为对照。结果:62例膀胱TCC组织标本中,EGFR mRNA在组织中平均阳性率72.6%(45/62)。在Tis-1、T2、T3、T4期膀胱TCC患者组织中EGFR mRNA阳性率分别为57.1%、62.5%、85.7%、100%;在Tis-1、T2、T3、T4期膀胱TCC患者外周血中EGFR mRNA阳性率分别为14.3%、25.0%、78.6%、90.9%。在组织和外周血中低分期(Tis-1、T2)与高分期(T3、T4)膀胱TCC表达EGFR mRNA阳性率差异有显著性(P<0.01)。在25例健康人外周静脉血中均未发现EGFR mRNA的表达。结论:膀胱TCC组织及外周血中EGFR mRNA表达阳性率与肿瘤分期、是否转移呈相关性。RT-PCR法检测膀胱TCC患者外周血EGFR mRNA有望成为检测膀胱肿瘤细胞循环内微转移的有效方法。
Objective: To explore the expression of epidermal growth factor receptor (EGFR) mRNA in peripheral blood of patients with bladder transitional cell cancer( TCC ) and its significance. Methods: Peripheral vein blood and tumor tissue samples of 62 patients with bladder TCC ( group TCC) were collected and peripheral blood of 25 healthy subjects were collected as negative control ( group C). The expression of EGFR mRNA was detected by RT-PCR. Results: The positive rates of EGFR mRNA expression averaged 72.6% (45/62) including 57.1% ,62.5% ,85.7% ,and 100 % in tissue samples and 14.3% ,25.0% ,78.6%, and 90.9% in peripheral blood samples from group TCC of stages of Tis-1, T2,33, and T4 respectively. There was a significant difference of EGFR mRNA expression in tissue and blood samples of TCC between stages Tis-1 ,T2 and stages T3, T4 ( P 〈 0. 01 ) . EGFR mRNA was not detected in the 25-healthy people. Conclusion: Positive rates of EGFR mRNA expression in samples of tissue and peripheral blood were correlated with the staging and grading, and metastasis of TCC. RT-PCR might be one of the effective methods for detecting micrometastasis in circulating of bladder tumor cells.
