摘要 目的:优选桑叶多糖的水提取工艺和观察桑叶多糖对小鼠免疫功能的影响.方法:采用正交试验方法,对影响多糖的水提取工艺的因素水平进行了研究,并采用腹腔注射环磷酰胺造模,测定桑叶多糖对免疫功能低下小鼠炭粒廓清、巨噬细胞吞噬功能,血清溶血素效价(HC50)、抗体细胞形成数量(QHS值)及T,B淋巴细胞功能的影响.结果:桑叶多糖的最佳提取工艺为1∶8倍量蒸馏水在80℃加热1.5h,回流提取1次;桑叶多糖能明显升高炭粒廓清k和a值以及HC50,QHS值(P<0.05~0.01),提高巨噬细胞吞噬百分率、吞噬指数和T,B淋巴细胞功能(P<0.01).结论:优选的桑叶多糖水提取工艺稳定可行,桑叶多糖具有增强小鼠免疫功能的作用. AIM:To optimize the water extraction techniques of polysaccharide from mulberry leaves and to explore the effect of mulberry leaf polysaccharide on the immune function of mice. METHODS: The conditions influencing the water extraction of polysaecharide were analyzed and optimized using the orthogonal test and the effects of the extracts on the carbon particle clearance coefficients, macrophage phagocytosis, serum hemolysin titer ( HC50), the amount of plaque forming cells ( quantitative hemolysis spectrophotometrically, QHS ), and functions of T-and B-lymphoeytes were studied in the mouse models of poor immune function induced by peritoneal injection of cyclophosphamide. RESULTS: The optimum conditions for the water extraction of polysaccharide from mulberry leaves were: the ratio of reagent to liquid was 1:8, heating at 80~C for 1.5 h and one-time recirculation. The extracts increased the k and a coefficients of carbon particle clearance, the levels of HC50 and QHS( P 〈 0.05 -0.01 ), the phagocytic rate and indexes of macrophages, and T-and B-lymphocyte functions ( P 〈 0. 01 ) in the mouse models. CONCLUSION : Our optimized extraction techniques for mulberry leaf polysaccharide are stable and feasible and the extracts enhance the immune function of mice.
