预处理对在体大鼠冠脉血管内皮细胞的保护及机制探讨 被引量:1

摘要 目的探讨缺血预处理(IPC)对缺血再灌注(I/R)所致的血管内皮细胞(VEC)损伤是否有保护作用及蛋白激酶C(PKC)在其中的作用。方法实验大鼠分为5组:对照组、缺血再灌注组(I/R组)、预处理组(IPC组)、多黏菌素B(PKC抑制剂)+I/R组、多黏菌素B+IPC组。测定各组一氧化氮(NO)、内皮素(ET)、循环内皮细胞(CEC)、丙二醛(MDA),并作冠脉VEC透射电镜观察。结果I/R后血浆中NO明显减少,ET明显升高,NO/ET值明显降低,CEC明显增多,MDA明显升高(P<0.05或P<0.01)。IPC能逆转上述改变,使血浆中NO增多,ET降低,NO/ET值恢复,CEC数量减少,MDA含量降低(P<0.05或P<0.01)。PKC的抑制剂多黏菌素B并不加重I/R的上述改变,但是能阻断IPC的作用。电镜显示:I/R后VEC的超微结构严重损伤,IPC能明显减轻VEC的结构损伤。结论I/R导致VEC结构和功能的严重损伤,IPC能减轻I/R后VEC的结构和功能损伤,PKC可能参与了这一过程。
