摘要
在组织病理学观察的基础上,经接种DF-1细胞系、ELISA抗原检测、聚合酶链式反应(PCR)以及亚群特异性免疫荧光试验(IFA),从送检的疑似禽白血病感染的麻黄肉种鸡中分离并鉴定出1株J亚群禽白血病病毒(ALV-J),命名为SCAU-0901。利用特异性引物分别扩增出长度为545 bp和2.2×103bp的目的片段,并对目的片段进行了克隆测序。序列分析发现,SCAU-0901株的gp85基因与国内外各ALV-J毒株相应核苷酸序列的相似性为86.1%~95.1%,其中与BJ0303株的相似性最高(95.1%),与ADOL-7501株的相似性最低(86.1%)。系统遗传进化分析表明,SCAU-0901株的gp85基因片段与BJ0303株的亲缘关系最近。
An subgroup J avian leukosis virus(ALV-J),designated as SCAU-0901,was isolated and identified by histopathological observation,DF-1 cell culture,ELISA test,PCR and immunofluorescence assay specific to ALV-J from Mahuang broiler breeder suspected with ALV infection.PCR amplification products,545 bp and 2.2×10 3 bp,were generated with two pairs of specific primers and sequenced.Accor-ding to the gp85 gene sequence comparison of SCAU-0901 strain with those of the other ALV-J strains at home and abroad,the sequences of gp85 gene of SCAU-0901 strain,shared 86.1% to 95.1% similarity with that of other ALV-J strains,the similarity with BJ0303 strain was the highest(95.1%),and the similarity with ADOL-7501 strain was the lowest(86.1%).Phylogenetic analysis showed that the gp85 gene of SCAU-0901 strain had the closest genetic relationship to BJ0303 strain.
