血管紧张素-(1-7)对大鼠肾系膜细胞TGF-β_1/Smad/CTGF通路的影响 被引量:1

摘要 目的探讨血管紧张素-(1-7)[Ang-(1-7)]对大鼠肾系膜细胞TGF-β1/Smad/CTGF通路的影响。方法采用WST法检测培养系膜细胞(GMC)增殖;RT-PCR法检测细胞内信号传递分子Smad3/7及结缔组织生长因子(CTGF)基因mRNA的表达。结果与对照组比较,Ang-(1-7)、明显抑制TGF-β1诱导的GMC增殖(P<0.05),同时,Ang-(1-7)对TGF-β1诱导的系膜细胞内Smad3、Smad7及CTGF基因mR-NA表达有明显的抑制作用(P<0.05)。结论Ang-(1-7)对TGF-β1诱导的系膜细胞增殖有抑制作用,其机制可能与抑制TGF-β1/Smad/CTGF通路有关。 [Objective] To investigate the effect of Angiotensin-(1-7) [Ang-(1-7)] on the TGF-β1/Smad/CTGF pathway of Cultured Glomerular Mesangial Cell. [Methods] Cell proliferation were evaluated by WST assay, and Smad3/7, CTGF mRNA expression in GMC were analyzed by RT-PCR. [ Results ] Compared with control groups, Ang-(1-7) obviously inhibit cultured GMC proliferation induced by TGF-β1 (P 〈0.05);and also inhibit the mRNA expression of Smad3/7and CTGF induced by TGF-β1 in cultured GMC (P 〈0.05). [Conclusion] Ang-(1-7) can inhibit the proliferation of GMC induced by TGF-β1 and the mechanisms may be associated with effects of suppressing TGF-β1/Smad/CTGF pathway.
