摘要
目的探讨异品系皮肤移植大鼠排斥反应发生过程中外周血γδT细胞的IFN-γ/IL-4分泌模式。方法建立Wistar→SD大鼠异品系皮肤移植模型,以SD大鼠自体皮肤再植为对照,于术后0、4、8d和10~13d(排斥时)采血,体外扩增外周血γδT细胞,用ELISA双抗夹心法测定培养上清和血浆中IFN-γ及IL-4的含量。结果异品系皮肤移植大鼠术后外周血γδT细胞培养上清中IFN-γ和IL-4水平均显著升高,且显著高于自体皮肤再植同时间点对照组;排斥反应发生时,血浆两种细胞因子含量和IFN-γ/IL-4比值均高于对照组;大鼠外周血γδT细胞培养上清IFN-γ、IL-4含量与血浆IFN-γ、IL-4含量分别呈正相关关系。结论异品系皮肤移植排斥反应发生过程中γδT细胞能分泌IFN-γ和IL-4,而γδT细胞IFN-γ/IL-4细胞因子的分泌模式可能影响血浆细胞因子格局,从而参与了移植免疫应答过程。
[ Objective ] To investigate the IFN-γ/IL74 cytokine pattern of γδT cell in rat allogenic skin transplantation model. [ Methods ] The rat allogeneic (Wistar to SD) skin transplantation model was established. Rat auto-skin replantafion was carried out as control group. Immobilized anti-TCRγδ monoclonal antibody (McAb) method was used to expand γδT cells from per/pheral blood in vitro. The concentrat/ons of IFN-γ and IL-4 in supernatants of γδT cells and in plasma were determined by ELISA. [ Results ] In allo-transplantation group, the levels of IFN-γ IL-4 and IFN-γ/IL-4 in supernatants and in plasma were significantly higher than in control group. The associations of cytokines concentrations between in supernatants and in plasma were significant in rat skin transplantation model. [Conclusion]γδT cells can produce IFN-γ and IL-4. The IFN-γ/IL-4 cytokine pattern of γδT cells partly influ- ences the pattern of plasma cytokines.
