益胃汤对初老雌性大鼠卵巢细胞凋亡线粒体通路的影响 被引量:6

Effect of Yiwei Decoction on Mitochondrion Path of Ovary Apoptosis of the Primary Senile Female Rats

摘要 目的探讨益胃汤延缓初老雌性大鼠卵巢机能衰老的机理。方法4~6月龄雌性SD大鼠为正常对照组;10~12月龄、阴道细胞学表现动情期延长的雌性SD大鼠作为初老大鼠模型,随机分为益胃汤高剂量组、益胃汤中剂量组、益胃汤低剂量组、己烯雌酚组、模型对照组。灌药4周后,取左侧卵巢用于检测卵巢Bcl-2、Bax、Caspase3、细胞色素C。结果与模型对照组比较,益胃汤可使卵巢Bcl-2表达增强,Bax表达减弱,Bcl-2/Bax比例增加(P<0.05),Caspase表达减少,细胞色素C减少,卵巢细胞凋亡减少,但达不到正常对照组水平(P>0.05),且益胃汤高、中、低剂量组在实验中呈现一定的量效关系。结论益胃汤通过抑制线粒体通路引起的级联反应抑制初老雌性大鼠卵巢细胞凋亡,这可能是其延缓卵巢机能衰老的机理之一。 Objective To explore the effect and mechanism of Yiwei Decoction(Decoction for reinforcing the stomach) in delaying the aging of the primary senile rat ovary.Methods The 4~6 months old SD female rats were set to be normal control group.The 10~12 months old SD female rats with vaginal cytological oestrus elongation were set as the primary senile female rat models,who were randomized into 5 groups:high dose Yiwei Decoction group;middle dose Yiwei Decoction group;low dose Yiwei Decoction group; diethylstilbestrol group; model control group. The intervention by gavage lasted 4 weeks. The left ovaries were tested by immunohistochemical method to observe the expression of Bcl-2, Bax, Caspase 3 and cytochrome C. Results Compared with the model control group, Yiwei Decoction could increase the expression of Bcl-2, decrease the expression of Bax, increase the proportion of Bcl-2/ Bax (P^0.05), decrease the expression of Caspase3, Cytochrome C, and ovary apoptosis, but not to the degree of the normal control group. And the high, middle and low dose Yiwei Decoction groups presented a dose-effect relationship. Conclusion The results showed that Yiwei Decoction can restrain ovary apoptosis through inhibiting the cascade reaction caused by mitochondrion path. This might be one of the mechanisms of Yiwei Decoction delaying the aging of ovary in primary senile rat .
