摘要
目的:考察糖芥苷的细胞毒性。方法:应用80%乙醇回流提取,正己烷萃取,MCI柱层析甲醇洗脱获得糖芥苷。采用体外培养的血管平滑肌细胞,通过检测细胞增殖、细胞内钙和c-myc基因的mRNA表达,考察糖芥苷细胞毒性,并与哇巴因、地高辛进行比较。结果:(1)糖芥苷浓度在10-5mg/mL时具有促血管平滑肌细胞增殖作用,增殖率为33.3%;哇巴因和地高辛浓度分别在10-14mg/mL和10-9mg/mL时具有促血管平滑肌细胞增殖作用,最大增殖率分别为49.08%和51.05%。(2)糖芥苷浓度在10-3mg/mL时可暂短升高细胞内钙;哇巴因浓度在10-4mg/mL时可持久升高细胞内钙。(3)糖芥苷浓度在10~8mg/mL时对c-myc mRNA表达没有影响;哇巴因和地高辛浓度在10-12mg/mL时可增加c-myc mRNA表达。结论:与哇巴因和地高辛相比,糖芥苷细胞毒性较小。
Objective:To investigate the cellular toxity of erysimosides. Methods:Erysimosides were obtained by extraction with 80% ethanol and isolation with MIC column. Toxic effects of erysimosides on proliferation, intracellular free calcium and c - myc mRNA expression in vascular smooth muscle cells were observed and compared with ouabain and digoxin. Results: ( 1 ) Erysimosides at the concentration of 10^-5mg/ml prese.nted the effect of stimulating proliferation; Ooabain and digoxin presented the effect of stimulating proliferation at the concentration of 10-14mg/ml and 10^-9mg/ml, respectively. (2) Erysimosides at the concentration of 10^-3mg/nil had a tentative increase of intracellular free calcium; Ouabain at the concentration of 10^-4 mg/ml increased appeared a permanentu of intracellular free calcium. (3) Erysimosides at the concentration of 10^-8mg/ml had no effect on c - myc mRNA expression; An increase of c - myc mRNA expression was observed by ouabain and digoxin at the concentration of 10 -12mg/ml, Conclusion:Compared with ouabain and digoxin, the toxity of erysimosides on vascular smooth cell is small.
