β-catenin和COX-2在大肠癌的表达及其相互作用

摘要 目的研究β-catenin和COX-2在大肠癌的表达及其意义。方法应用原位杂交和免疫组化法检测β-catenin和COX-2在正常黏膜、非瘤性息肉、腺瘤和大肠癌等4组不同大肠病变组织中的表达情况,分析其相互关系。结果β-catenin mRNA和蛋白以及COX-2蛋白在大肠癌的发生过程中均有异常表达,β-catenin mRNA阳性强度与β-catenin蛋白异位表达呈正相关,β-catenin蛋白的异位表达与COX-2蛋白阳性表达呈正相关。结论β-cate-nin和COX-2在腺瘤和大肠癌均有高表达,在大肠癌的发病过程中可能有一定作用。 Objective To investigate the abnormal expressions of β-catenin and COX-2 in colorectal carcinoma. Methods Checking the expressions of β-catenin mRNA within situhybridization, β-catenin and COX-2 protein with immunohistochemistry in normal colorectal mucosa, non-tumorous polyps, adeno- mas and adenocarcinomas, and analyzing their interacting relationship. Results The expression of β- catenin in mRNA/protein and COX-2 protein was higher in the generation and development of colorectal cancer, meanwhile the high expressions of β-catenin mRNA were positively correlated with the heterotopical expression of β-catenin protein and the heterotopical expression of β-catenin protein was positively correlated with the positive expressions of COX-2 protein at the same time. Conclusion There are high expressions of β-catenin in mRNA/protein and COX-2 protein in adenomas and adenocarcinoma, and their interaction with each other makes carcinogenesis ultimately.
