Castleman′s病患者的CD4^+CD25^+调节性T细胞的研究

Study on Changes of CD4^+CD25^+ T Regulatory Cells in Peripheral Blood of Patients with Castleman's Disease

摘要 目的:探讨Castleman′s病患者外周血中CD4+CD25+调节性T细胞的变化及其与疾病的关系。方法:用流式细胞术对14例Castleman′s病患者及30例正常对照的外周血中CD4+CD25lowT细胞、CD4+CD25highT细胞、CD4+CD25highFOXP3+T细胞的比例进行检测。结果:局灶型Castleman′s病患者外周血中CD4+CD25lowT细胞、CD4+CD25highT细胞和CD4+CD25highFOXP3+T细胞比例在治疗前分别为(4.55±0.53)%、(0.73±0.09)%、(0.64±0.09)%;治疗后分别为(4.88±0.24)%、(1.0±0.10)%、(0.93±0.10)%,均低于正常对照组。治疗后这3种T细胞的比例较治疗前有显著升高(P<0.05)。多中心型患者外周血中这3种T细胞比例在治疗前分别为(9.43±0.81)%、(2.79±0.17)%、(2.46±0.09)%;治疗后(8.82±1.01)%、(2.71±0.10)%、(2.23±0.10)%,均高于正常对照组。结论:CD4+CD25+调节性T细胞在局灶型和多中心型Castleman′s病中的变化是不同的,这种不同的变化可能和这两型的临床表现和预后不同有关。 Objective:To investigate the changes of CD4^+CD25^+ T regulatory cells in peripheral blood of Castleman s disease(CD) and the correlationship with the disease.Methods:The proportion of CD4^+CD25^low,CD4^+CD25^high and CD4^+CD25^high FOXP3+ T cells in peripheral blood were measured in 14 patients with Castleman s disease before and after treatment and in 30 normal controls by flow cytometry.Results:The percentage of CD4^+CD25^low,CD4^+CD25^high and CD4^+CD25^highFOXP3+ T cells in localized CD were(4.55±0.53)%,(0.73±0.09)% and (0.64+0.09) % respectively, before treatment ,significantly lower than the corresponding values in the controls,and was (4.88± 0.24)%, (1.03 ±0. 10)% and (0. 93±0. 10)% respectively,after treatment,demonstrating the increase of the 3 subtypes of T cells in peripheral blood (P〈0. 05). The percentage of CD4^+CD25^low,CD4^+CD25^highand CD4^+CD25^highFOXP3^+T cells in multicentric CD were higher than those in the controls, and were (9. 43±0.81)%, (2. 79±0.17)% and (2. 46±0.09)% respectively, before treatment, and (8.82 ± 1.01) %, (3.71 ± 0. 10) % and (2.23 ± 0.10) % ,respectively,after treatment, significantly higher than the corresponding values in the controls. Conclusion:In localized type and rnulticentric type of CD, the changes of CD4^+CD25^+T regulatory cells are quite different, probably relating to the divergence of the two types in clinical manifestations and prognosis.
