摘要
目的研究人食管鳞状细胞癌与癌旁正常组织的基因表达谱,通过对比基因表达的差异,寻找与人食管鳞癌发展相关的基因。方法提取人食管鳞癌与正常食管鳞状上皮的mRNA,逆转录成cDNA,并用Cy3-dUTP标记正常食管组织,用Cy5-dUTP标记食管鳞癌组织,制成探针,与基因芯片进行杂交,经扫描仪扫描后,对获取的信号数据进行软件分析、对比,筛选出差异表达的基因。结果在4例新鲜标本的芯片杂交检测中,共发现4329条基因发生差异表达,其中2293条上调,2036条下调,将这些基因按GO(gene ontology)分子功能(function term)进行分类,发现与细胞分化、成熟,分子定位、连接,信号传导,酶活性调节,以及基因转录、翻译调节有关的基因最多。结论①食管鳞癌的发生发展与多条基因的异常表达有关。②应用基因芯片技术可以对食管鳞癌的基因表达谱进行分析,以筛选食管鳞癌相关基因。
Objective To investigate gene expression in human esophageal squamous cell carcinoma and adjacent normal tissue, and to find the genes that were relevant to the genesis and development of human esophageal squamous cell carcinoma by comparing gene expression differences. Methods Total mRNA was extracted from human esophageal squamous cell carcinoma and adjacent normal tissue and cDNA was acquired by reverse transcription reaction. Probe was made by cDNA which labeled with Cy3-dUTP from normal esophageal tissues and labeled with Cy5-dUTP from esophageal carcinoma, then hybridized with gene chip, scanned, the signal data acquired by software analysis and comparison, selected out genes different in expression. Results In four cases of fresh specimens hybridizaed with gene chip, a total of 4 329 different in gene expression was found, including 2 293 up-regulation,2 036 down-regulation. These genes were divided by molecular function(function term) according to GO ( gene ontology) Category, the most of these genes are relevant to cell differentiation, maturation, molecular orientation, connectivity, signal transduction, enzyme activity regulation, as well as gene transcription and translation. Conclusion ①Many genes that expressed abnormally played role in genesis and development of esophageal squamous cell carcinoma; ②Gene chip technology could be used to analyze gene expression profiles of esophageal cancer and select out genes related to esophageal squamous cell carcinoma.
