红斑狼疮患者血清对IL-6报告基因激活的研究

Effect of Serum of Patients with Active Systemic Lupus Erythematous on Expression of Interleukin-6 Reporter

目的构建白介素-6报告基因(IL-6reporter)载体并研究活动期系统性红斑狼疮(Systemic Lupus Erythema-tosus,SLE)患者血清对IL-6表达的影响。方法利用标准的分子克隆技术方法将IL-6的启动子区(-590～-1)重组到pGL3-basic载体中得到IL-6reporter,然后转染细胞并研究活动期系统性红斑狼疮患者血清对报告基因活性的影响。结果IL-6reporter的活性达到空载体的5倍左右,而活动期系统性红斑狼疮患者血清能使IL-6reporter的活性进一步升高(1.43±0.53)倍,而正常对照的血清对IL-6reporter无明显的增强作用。结论在活动期系统性红斑狼疮患者血清中存在有促进细胞表达IL-6的因素存在。

Objective To construct IL-6 reporter and study the effect of serum prepared from patients with active Systemic Lupus Erythematous（SLE） on the expression of reporter. Methods IL-6 reporter was constructed by cloning IL-6 promoter [ （ - 590） -（ - 1 ） ] into pGL3-basic vector and used to transfect culture cells;The effect of serum of patients with active SLE on the re- porter expression was observed. Results The activity of IL-6 reporter was about 5 times that of pGL3-basic, and the serums from patients with active SLE further promoted the activity of reporter（ 1.43 ± 0. 53 ） times, while serums from normal control had no obvious effect. Conclusion Some factors in the serums from patients with active SLE can promote cells to express the IL-6.
