摘要
目的观察不同浓度的臭氧在不同作用时间对家兔椎间盘细胞的作用影响。方法首先建立家兔椎间盘细胞的体外培养系统,其中,第一组给予纯氧,另外3组给予不同浓度的臭氧:30μg/ml,60μg/ml和80μg/ml,然后于15min及30min后,采用台盼蓝细胞排斥试验计算椎间盘细胞的活细胞率;结果低浓度组活细胞率为80%左右,高浓度组活细胞率10%左右。15min时,不同浓度组之间台盼蓝细胞排斥试验结果差异都具有统计学意义;30min时,对照组与低浓度组差异无统计学意义,其他组之间存在统计学意义;不同时间只有中浓度组存在统计学意义。结论随着臭氧浓度的增加,作用时间的延长,椎间盘细胞活细胞率呈降低趋势。
Objective To observe the effectof O2-O3 mixture of different concentration at different time- points on intervertebral disc cell cultured in vitro. Methods Monolayer culture cells were divided to 4 groups. The first group was given pure 02 ,and other groups were given different concentrations of 03:30 μg/ml,60 tLg/ml and 80μg/ml, trypan blue test was donel5 and 30 minutes afterwards. Results Cell viability was about 80% in 30 μg/ml group, and 10% in 80μg/ml group. After 15 minutes, trypan blue test showed significant differents in each group. After 30 minutes, there's no difference of result of trypan blue test between control group and 30μg/ml group, while there's difference among control group, 60 μg/ml group and 80 μg/ml group. There's significant difference only in 60μg/ml group at 15 and 30 minutes. Concision With concentration and duration of 03 increasing, number of live intervertebral disc cell decreases.
