IGF信号通路和雄激素受体在前列腺癌发展过程中的相互作用 被引量:1

摘要 胰岛素样生长因子I受体(IGF—IR)目前被认为在前列腺癌发展过程中扮演了一个很重要的角色。很可能导致了雄激素非依赖性前列腺癌(AIPC)的发生。雄激素受体(AR)的表达和雄激素调控基因是导致激素阻断治疗后前列腺癌进展的根本原因。因此,生长因子的信号途径被认为在激素阻断后,激素依赖性前列腺癌转变成AIPC过程中发挥了一定的作用。阻断IGF—IR信号通路可以维持细胞质中的AR并且导致雄激素调控基因表达的显著改变。而AR从细胞质中转移到细胞核中可能和IGF介导的AR的磷酸化有关。目前已经有多种以IGF—IR为靶点的抗体应用于临床研究。本文将分析前列腺癌细胞中IGF—IR信号通路如何调节AR结构,从而改变AR活性的研究进展,为以IGF—IR为靶点的治疗AIPC的前景作一综述。
