摘要
目的研究帕金森病(PD)黑质多巴胺神经元的死亡机理。方法用脱氧核糖核酸(DNA)标记法,借助流式细胞仪和荧光显微镜观察多巴胺(DA)对大鼠嗜铬细胞瘤(PC12)细胞凋亡的影响。结果DA可诱发PC12细胞凋亡,适中浓度(0.45mmol/L)时PC12细胞凋亡数最多(凋亡率为48.7%±6.3%);抗氧化剂二硫苏糖醇(DTT)可部分阻止DA诱发的PC12细胞凋亡(P<0.01)。结论细胞凋亡参与了PD的病变过程。
Objective To study the mechanism of dopaminergic neuronal death in substantia nigra of Parkinson's disease (PD). Methods Using the DNA endlabeling method the effect of dopamine (DA) on rat apoptotic PC12 cells was examined by flow cytometry and fluorescence microscopy. Results DA induced apoptosis in PC12 cells. Moderate concentration of DA (045mmol/L) gave maximal number of apoptotic cells with a rate of apoptosis 487%±63%; and the antoxidant dithiothreitol (DTT) partially blocked DAinduced apoptosis in PC12 cells (P<001). Conclusions Apoptosis may be involved in the pathogenesis of PD, and treatment with proper antioxidants may be useful in PD therapy.
