CD14启动子-159位点基因多态性与糖尿病肾病的相关性研究 被引量:5

摘要 目的研究2型糖尿病患者CD14启动子C-159T多态性分布,探讨该基因多态性与糖尿病肾病(ON)的相关性。方法应用聚合酶链反应-限制性片段长度多态性技术对308例2型糖尿病患者及145名正常对照者CD14基因启动子-159位点进行基因型分析。结果(1)CD14启动子-159位点基因多态性在正常对照组和2型糖尿病组中的分布差异无统计学意义(P〉0.05);在未发生DN和发生DN的患者中,CD14启动子-159位点CC基因频率与CT+TT基因频率相比,差异有统计学意义(P〈0.05);(2)2型糖尿病患者在10年和20年时,发生DN的百分率分别为13.3%和23.4%;纯合子CC基因型2型糖尿病患者发生DN的风险高于CT+TT基因型患者。结论CD14启动子C-159T基因多态性与糖尿病的发病无关,但其中的CC纯合子基因是2型糖尿病患者进展为DN的遗传学风险因素。 Objective To investigate the distribution of CD14 promoter gene C-159T polymorphsim in patients with type 2 diabetes mellitus (T2DM) and to analyze the association of CD14 polymorphisms with diabetic nephropathy (DN). Methods Genotypes of CD14 were typed in 308 T2DM patients and 145 normal controls by polymerase chain reaction restriction fragment length polymorphism. Results (1)The distribution of CD14 promoter gene C-159T polymorphsim revealed no significant difference between normal control group and T2DM group(P〉0.05). The distributions between CC genotype and CT+TT genotypes existed statistically significant difference in CD14-159 between two subgroups with and without diabetic nephropathy (P〈0.05). (2)At 10 and 20 years after the onset of DM, 13.3% and 23.4% of the patients developed into DN respectirely. CC genotype had higher risk than CT+TT genotypes to develop into DN. Conclusion The distribution of CD14 promoter gene C -159T polymorphism has no relation with T2DM. The CC genotype of the C-159T polymorphism of CD14 gene is a risk factor for diabetic nephropathy.
