摘要 目的研究重组腺相关病毒介导的CD151基因转染人脐静脉内皮细胞(HUVECs)对整合素α3β1/α6β1表达的影响。方法包装携带CD151、CD151-QRD和CD151-ARSA的重组腺相关病毒,转染HUVECs细胞,用Western Blot和流式细胞仪检测HUVECs细胞CD151及整合素a3131/a6131的表达。结果CD-51组及其各突变体组的CD151表达均明显高于对照组(P〈0.01),但3组间互相比较CD151蛋白表达差异无统计学意义(P〉0.05)。Western Blot及流式细胞仪检测显示CDl51组α3、α6及β1的表达明显高于对照组、CD151-QRD组和CD151-ARSA组(P〈0.05),其余各组间比较差异无统计学意义(P〉0.05)。结论CD151基因转染能促进整合素α3β1/α6β1的蛋白表达,其机制可能与CD151胞外大环QRD194-196。“及C末端囊泡运输基序YRSL”。。。有关。 Objective To investigate the effects of recombinant adeno-associated virus (rAAV)-CD151 on the expression of the integrin α3β1/α6β1 in human umbilical vein endothelial cells (HUVECs). Methods rAAV-CD151, rAAV-CD151-QRD and rAAV-CD151- ARSA were produced and transfected into HUVECs directly. Protein levels of CD151 were detected. And the protein expression of integrin α3 ,α6 and β1 on the surface of HUVECs were measured by western blot and flow cytometry. Results Transfections of rAAV-CD151 increased the expression of CD151 ( P 〈0. 01 )and integrin α3β1 /α6β1 ( P 〈 0. 05). On the contrary, transfections of rAAV-CDI51-QRD and rAAV-CD151-ARSA only increased expression of CD151 ( P 〈0. 05), but didn! increase expression of integrin α3β1 /α6β1. Conclu-sion CDI51 gene transfection could promote the expression of integrin oL3131/ct6131. The mechanism of regulating integrin α3β1/α6β1 may lie in allosterism of an extracellullar CD151 site (QRD194-196) and C -terminal Ves. transportation target motif(YRSL245-248).
