蛇毒金属蛋白酶抑制剂对小鼠黑色素转移瘤血管生成拟态形成的影响 被引量:3

摘要 目的:研究蛇毒金属蛋白酶抑制剂BJ46a对血管生成拟态形成的影响。方法:将BJ46a基因转染B16细胞,建立稳定转染的B16/pcDNA3.1HisC-BJ46a细胞株,通过C57BL/6小鼠实验性肺转移模型,模拟肿瘤细胞降解局部细胞外基质、进而侵袭转移过程,应用光镜和超薄切片透射电镜技术观察BJ46a基因转染对血管生成拟态形成的影响。结果:小鼠黑色素瘤内存在血管生成拟态,稳定转染的B16/pcDNA3.1HisC—BJ46a细胞株抑制其肿瘤血管生成拟态的形成。结论:BJ46a通过抑制血管生成拟态形成,从而抑制肿瘤的侵袭转移。 Objective: To study the influence of snake venom metalloproteinase inhibitor BJ46a on mouse melanoma vasculogenic mimicry. Methods: BJ46a gene was transfected into the murine melanoma cell line B16, and then stable transfection BJ46a clones (B16/pcDNA3.1HisC-BJ46a) were obstained. The ability of tumor cell invasion and migration was identified by tumor invasion animal model in vivo. Results: Animal experiment and electron microscopy proved the existence of mouse melanoma vasculogenic mimicry. In control group, neoplastic cells were surrounded to form tube where there were erythrocytes. And this structure was seldom seen in B16/pcDNA3.1 HisC-BJ46a group. Conclusion: Transfection of BJ46a gene into mouse melanoma cell line results in the inhibition of vasculogenic mimicry formation, accordingly suppress tumor invasion and metastasis.
