摘要
目的:观察左旋多巴对大鼠脑内5羟色胺(5-HT)能神经元的影响。方法:左旋多巴大鼠灌胃,以免疫细胞化学和流式细胞术显示5-HT及色氨酸羟化酶(TPH)免疫反应物质或含量的变化。结果:免疫细胞化学显示左旋多巴组大鼠中缝背核5-HT能神经元细胞直径明显小于对照组;中缝背核5-HT和TPH免疫反应物质灰度值比对照组分别增高了7.72%和16.57%。流式细胞术显示左旋多巴组5-HT的表达量比对照组减少了18.45%,TPH表达量比对照组减少了19.90%。结论:左旋多巴降低脑内5-HT能神经元5-HT和TPH的表达。
Objective: To study the effect of L-dopa on the serotonergic neurons in the rat brain. Methods: Wistar rats were treated with L-dopa by intragastric administration. Immunocytochemistry and flow cytometry were mainly used to analyze the altered expression of serotonin (5-HT) and tryptophan hydroxylase (TPH) in L-dopa-treated rats. Results: Immunohistochemistry showed that the diameter of 5-HT neurons in dorsal raphe nucleus (DRN) was smaller than that in control and the mean gray value of 5-HT and TPH in DRN of the L-dopatreated rats increased by 7.72% and 16.57% respectively. Flow cytometry showed that the expression of 5-HT and TPH in DRN of the L-dopa-treated rats significantly decreased by 18. 45 % and 19. 90% respectively compared with the corresponding control. Conclusion: L-dopa decreases the expression of 5-HT and TPH in the rat brain.
