紫杉醇联合放疗治疗鼻咽癌裸鼠移植瘤过程中Survivin mRNA的表达及其意义 被引量:6

Expression and significance of Survivin mRNA in xenotransplanted nasopharyngeal carcinoma treated by paclitaxel combined with radiotherapy

摘要 目的:研究紫杉醇联合放疗治疗鼻咽癌CNE-2裸鼠移植瘤的疗效及凋亡抑制基因Survivin的表达和意义。方法:建立鼻咽癌CNE-2裸鼠移植瘤,分别进行紫杉醇(紫杉醇组)、放疗(放疗组)、紫杉醇+放疗(紫杉醇+放疗组)处理,测量移植瘤体积并对移植瘤标本进行苏木精-伊红染色、流式细胞仪检测凋亡指数及one step RT-PCR检测Survivin mRNA的表达。结果:紫杉醇+放疗组对裸鼠移植瘤的生长抑制最为明显,抑制率为99.3%。与对照组相比,紫杉醇组、放疗组和紫杉醇+放疗组的凋亡指数均明显增加(P<0.05),其中紫杉醇+放疗组更为明显(P<0.05)。紫杉醇组和放疗组的裸鼠移植瘤组织中Survivin mRNA的表达与对照组无明显差异(P>0.05),但紫杉醇+放疗组中Survivin mRNA的表达显著下降(P<0.05)。结论:紫杉醇联合放疗对鼻咽癌低分化移植瘤具有明显的杀伤作用,紫杉醇对鼻咽癌裸鼠移植瘤具有放疗增敏作用,这种作用可能与凋亡抑制基因Survivin的表达下调有关。 Objective: To investigate the expression and significance of Survivin mRNA in xenotransplanted nasopharyngeal carcinoma treated by paclitaxel combined with radiotherapy. Method: Xenotransplanted nasopharyngeal carcinoma was established by CNE 2 cell line, then grouped and treated with paclitaxel, radiotherapy, paclilaxel combined with radiotherapy respectively. Xenotransplanted tumor volume was measured; tumor specimens were comfirmed by routine hemotoxylin-eosin staining; apoptosis index was assayed by flow cytometry and Survivin mRNA was detected by one step RT-PCR. Result: Xenotransplanted tumor growth was significantly inhibited by paclitaxel combined with radiotherapy and its inhibition rate was 99.3 %. Compared to control group, apoptosis index was apparendy increased in the other three groups(P〈0. 05), especially in the combined therapy group (P〈0. 05). Survivin mRNA expression was obviously decreased in the combined therapy group (P〈0. 05) whereas there was no difference in its expression among the groups of paclitaxel, radiotherapy,and control group (P〉0. 05). Conclusion: Paclitaxelcombined with radiotherapy can induce significant killing effect in xenotransplanted nasopbaryngeal carcinoma; paclitaxel can enhance the radiosensitivity of xenotransplanted nasopharyngeal carcinoma and its mechanism may rely on the down-regulation of Survivin expression.
