摘要
目的观察白藜芦醇对帕金森病大鼠的保护作用,并探讨其可能机制。方法应用6-OHDA纹状体内注射制作PD模型大鼠,将模型成功大鼠随机分为4组:模型组及白藜芦醇(10,20,40mg/kg/d)治疗组,并设假手术组及正常给药组(白藜芦醇20mg/kg/d),每日口服给药一次,连续10周,给药过程中每周进行一次行为学检测。电镜下观察黑质神经元超微结构改变,RT-PCR法检测大鼠黑质部caspase-3mRNA的表达。结果①白藜芦醇能改善大鼠的旋转行为;②白藜芦醇能减轻6-OHDA诱导的帕金森病大鼠黑质多巴胺能神经元的变性;③白藜芦醇抑制大鼠黑质中caspase-3mRNA的表达。结论白藜芦醇对帕金森病大鼠产生保护作用,其机制可能与caspase-3mRNA的表达下调有关。
Objective To observe the protective effect of resveratrol (Res) on Parkinson's disease (PD) rats, and explore its potential mechanism. Methods The PD model rats were induced by injecting 6-OHDA into the right striatum in rats, The valid PD rats were randomly divided into four groups: model control group, resveratrol groups (10, 20 and 40 mg/kg/d), sham group and normal administration group (resveratrol 20 mg/kg/d). Res was given orally once daily for 10 weeks, Rotational test was examined weekly during administration. The ultrastructure of suhstantia nigra tissues was observed by electronic microscope, the levels of caspasc-3 mRNA was determined by RT-PCR.Results ①Rcs could decrease the turns of PD rats. ②Res could alleviate 6-OHDA induced degeneration of dopaminergic neurons in rat substantia nigra. ③Res could decrease the expression of caspase-3 mRNA in rat substantia nigra. Conclusion Res have protective effect on PD model rats, which may be related to down regulation of caspase-3 mRNA.
