摘要
目的:筛选适合分离和纯化荔枝核总黄酮的大孔吸附树脂并确立纯化工艺参数。方法:以吸附及解吸率为指标考察8种型号的树脂,确定纯化荔枝核黄酮的最佳树脂。并通过单因素分析考察该树脂分离、纯化荔枝核总黄酮的最佳工艺条件。结果:D4006吸附率及解吸率均高于其他7种树脂。其具体工艺条件为控制上样浓度5.45mg/mL、流速0.5mL/min,上样体积4BV,洗脱剂浓度为80%,洗脱剂用量14BV。结论:D4006型树脂在所确定的工艺条件下,纯化荔枝核总黄酮效果良好,总黄酮得率可达近90%。
Objective : To select the suitable resin and establish the optimal conditions for separation and purification of total flavone from Semen Litchi. Methods : To determine the optimalone by inspection of four types ofmacroporous resin. The optimal process of purification was studied by single factor analysis. Resuhs : The static absorption capacity, the absorption ratio and the static elution ratio of D4006resin was higher than the others. The optimal technological conditions were found in D4006 as follows: sample concentration was 5. 45 mg/mL, flow rate was 0. 5 mL/min, sample volume was 4 BV, and 80% ethanol was used as elufing agent and its dosage was 14 BV. Conclusion : The type of D4006 showed comprehensive property in the condition and the yield of the total flavones achieved nearly 90%.
