色素上皮源性因子在前列腺增生组织中的表达

摘要 目的观察色素上皮源性因子(PEDF)在良性前列腺增生组织中的表达,探讨这种血管源性因子在前列腺良性增生性疾病发生、发展中的作用。方法以临床收集到的确诊为前列腺增生病变37例组织及来自意外死亡成年男性志愿者的正常前列腺组织7例标本作为研究对象。以HE染色观察病理状况下前列腺的组织结构改变;以免疫组织化学染色法定位并定量PEDF在前列腺组织中的表达,并比较两组间表达的差异性。结果 HE染色显示前列腺增生病变表现为特征性的组织病理学改变。免疫组织化学染色观察到PEDF表达主要定位于前列腺组织的腺上皮细胞、基质细胞,尤其是平滑肌细胞;在良性前列腺增生组织中PEDF表达逐渐减少,两组间比较差异有显著性(P<0.05)。结论正常前列腺组织有PEDF的大量表达,表明其在维持正常机体前列腺的完整性方面发挥一定的作用;前列腺增生组织PEDF表达较正常明显减少,提示减少的血管形成抑制因子可能促进了这种疾病的发生、发展。 Objective To observe the expression of pigment epithelium derived factor(PEDF) on benigh prostate hyperplasia (BPH) ,investigate its function and the possible mechanism of neovascularization in prostate diseases. Methods The prostate tissue from thirty-seven patients who were diagnosed as BPH definitely and seven male donors who died unexpectedly were collected. The histopathological changes of the tissue was observed by using hematoxylin eosin method,and the expression of PEDF using immunohistoehemical staining. Results The specimens of BPH had its characteristic histopathologieal changes. The expression of PEDF was mainly localized in prostate epithelial cells and stromal cells, especially in smooth muscle cells. In normal donors'prostate tissue, PEDF was highly expressed; Whereas in BPH group, it was decreased. There was significant difference between the two groups( P 〈 0.05 ). Conclusion PEDF in normal prostate tissue may play physiological role in maintaining the integrity and functions of prostate. The decreased expression of PEDF in BPH tissue may have inducible roles in disease aggravation.
