摘要
目的:探讨复方黄连降糖片治疗2型糖尿病的可能作用机制。方法:对链脲佐菌素注射加高脂饮食诱导的2型糖尿病模型大鼠,给予复方黄连降糖片治疗8周。氧化酶法测定血糖,放射免疫双抗体法测定血清胰岛素,RT-PCR法测定大鼠肝脏、骨骼肌和脂肪组织IRS-1 mRNA的表达及Western blot法测定肝脏、骨骼肌和脂肪组织IRS-1蛋白的表达。结果:复方黄连降糖片治疗组大鼠的血糖和血清胰岛素均有所下降,各组织IRS-1的基因表达均显著提高。结论:复方黄连降糖片可能通过提高外周组织IRS-1的基因表达,从而对2型糖尿病起到治疗的作用。
Objective: To investigate the possible mechanism of "Compound Huanglian Jiangtang Tablet" (CHJT) in treating type 2 diabetes. Methods: The model of type 2 diabetic rats was induced by streptozotocin injection and high-fat diet and administered with CHJT for 8 weeks. Blood glucose was detected by enzymic method, blood insulin was detected by RIA, the expression of IRS-1 mRNA and protein in liver, muscle and fat of type 2 diabetic rats were determined by RT-PCR and western blot analysis. Results: In CHJT group, both the level of blood glucose and blood insulin decreased, and the mRNA and protein expressions of IRS-1 in liver, muscle and fat tissue increased significantly. Condusion:CHJT may treat the type 2 diabetes by raising the IRS-1 mRNA and protein expression in peripheral tissue.
