摘要
目的:观察三七总皂甙(PNS)对血清和肺组织血管紧张素转换酶(ACE)的影响。方法:①PNS与马尿酰甘氨酰甘氨酸和血清或肺组织匀浆共浴;②静脉注射PNS,应用紫外分光光度法测定马尿酸产出率,以其代表ACE活性。结果:离体反应显示,随着PNS浓度增高(0.15~1.2mg/ml),大白鼠血清、肺组织和人血清ACE活性逐渐下降(P<0.01)。PNS为1.2mg/ml时,抑制率分别为(61.54±11.76)%、(72.54±7.83)%和(62.09±16.4)%,三者之间无显著性差异(P>0.05)。PNS(75mg/kg)静脉注射家兔血清ACE活性从(62.52±12.43)IU下降为(44.80±10.47)IU(P<0.001)。结论:PNS对循环和局部组织ACE活性均有抑制作用。
Objective:To investigate the effect of panax notoginseng saponins (PNS) on angiotensin converting enzyme (ACE) activity Methods:①PNS, hippuryl glycylglycing and serum or lung tissue homogenate were mixed and incubated in vitro ②PNS was injected intravenously The ACE activity was determined by measuringthe hippuric acid generated from hippuryl glycylglycing, the substrate, with a UV spectrophotometer Results: PNS at 0 15 ̄1 2 mg/ml inhibited the ACE activity in a concentration dependent manner The inhibition of ACE by PNS at 1 2 mg/ml in the rat serum, the rat lung tissue homogenate and the human serum were (61 54±11 76)%,(72 54±7 83)% and (62 09±16 43)%, respectively It is no significient difference between these three (P>0 05) At 75 mg/kg, PNS decreased the sernm ACE activity in rabbit from (62 52±12 43)IU to (44 80±10 47)IU Conclusion: PNS has an inhibition effect on the activities of ACE in circulation and local tissue The mechanism of the inhibition effect of PNS needs to be futher investigated and might be defferent from that of the synthetic ACE inhibitors
