摘要
目的用体外系统血液固有免疫反应体系研究银屑病患者红细胞固有免疫功能状况,并探讨其在银屑病发病机制中的作用。方法将灭活大肠杆菌悬液(3×10^8/ml)0.2ml作为免疫原激活剂加入枸橼酸抗凝的全血细胞悬液0.2ml和血浆0.3ml中,37℃水浴1h,用流式细胞仪测定红细胞CD59表达量。结果加入大肠杆菌的银屑病全血细胞组(银屑病实验组)红细胞CD59的表达量(749.27±151.33)高于银屑病对照组红细胞CD59的表达量(651.44±152.35)(P〈0.05),明显高于加入大肠杆菌的健康试验组红细胞CD59的表达量(533.61±171.99)(P〈0.01);银屑病对照组红细胞CD59的表达量(651.44±152.35)较健康对照组红细胞CD59的表达量(475.17±124.97)明显升高(P〈0.01);银屑病患者红细胞CD59的表达量与病情严重程度指数评分呈正相关(r=0.726,P〈0.01)。结论银屑病患者红细胞固有免疫功能亢进,在银屑病免疫发病机制中可能起重要作用;银屑病患者红细胞固有免疫功能的测定可作为判断银屑病病情的客观指标之一;银屑病患者红细胞对外来抗原反应调节机制呈现紊乱状态。
To study the red blood cell(RBC) innate immune function in patients with psoriasis, and to find its possible role in the pathogenesis of psoriasis. Methods E. col: suspension (3×10^8/ml)0.2 ml was mixed with 0.2 ml whole blood cells treated by citric acid and 0.3 ml plasma. The mixture was incu- bated for lh at 37℃. The expression level of CD59 in red blood cells was determined by Flow Cytometry. Results The expression level of CD59 (749.27 ± 151.33 ) in red blood cells treated with E. coli was higher than that of CD59(651.44 ± 152.35)in red blood cells treated with isotonic saline(P 〈 0.05 )and CD59 (533.61 ± 171.99)in red blood cells of treated with E. coil. (P 〈 0.01 ). The expression level of CD59 (651.44 ± 152.35 ) in psoriasis control group was significantly higher than that of CD59 (475.17 ± 124.97 ) in normal control group( P 〈 0.01 ). The expression level of CD59 on red blood cells of psoriasis was positively correlated with the PASI ( r = 0.726,P 〈 0.01 ). Conclusion The innate immunity hyperfunction of red blood cells in patients with psoriasis may play an important role in the immune pathogenesis of psoriasis. The determination of the innate immune function of red blood cells in psoriasis patients could likely be used as one of objective indicators for the evaluation of the disease. The adjustment mechanism of red blood cells in psoriasis is in a disordered state.
