三种血虚证动物模型的建立及造血损伤的变化特点 被引量:11

摘要 目的系统考察^(60)Coγ射线、环磷酰胺和综合放血法致血虚证发生发展的特点,为全面评价中医血虚证模型的病变规律提供理论依据。方法采用^(60)Coγ射线照射、环磷酰胺腹腔注射和综合放血法制作血虚证动物模型,检测外周血各血液成份、骨髓造血干/祖细胞、CD34+细胞等指标变化。结果3种血虚证小鼠外周血中的主要血液成分下降,骨髓造血干/祖细胞的集落生成能力降低,骨髓中CD34+细胞减少。但从血虚证发生发展的过程来看,3种血虚证小鼠又存在不同特点:辐射损伤法主要影响骨髓造血功能,环磷酰胺致血虚证小鼠白细胞变化明显,综合放血法直接造成外周血中的红细胞和血红蛋白数量减少。结论3种模型均符合血虚证表现,但血虚证发生发展各有特点。 Aim To investigate factors which play important roles in the mechanism of pathogenesis and development of blood deficiency(BD)induced by 60Co irradiation,cyclophosphamide(CTX)or chronical bleeding,and to evaluate characters of BD.Methods Three BD animal models were induced by 60Co irradiation,CTX or chronical bleeding.The number of circulating blood cells,the ability of colony-forming unit of bone marrow cells,and the percentage of CD34+ cells in bone marrow were observed.Results The number of circulating blood cells,the ability of colony-forming unit of bone marrow cells,and the percentage of CD34+ cells in the BD animals decreased.Three BD animals had different characters.The hematopoietic function in bone marrow was reduced evidently in 60Co irradiation-induced BD model;white blood cells changed remarkably in CTX model and the number of red blood cells and hemoglobin decreased in the chronically bleeding model.Conclusion Three BD animal models had characters of BD,and of different nature in pathogenesis and development of BD.
