在乳腺癌细胞MCF-7中紫杉醇诱导硫氧还蛋白结合蛋白-2的表达

摘要 探讨紫杉醇在人乳腺癌细胞株MCF-7中诱导硫氧还蛋白结合蛋白-2表达及与其诱导的相关途径.用不同浓度的紫杉醇刺激MCF-7细胞,MCF-7细胞生长受抑制.通过荧光实时定量PCR检测,发现紫杉醇刺激MCF-7细胞后硫氧还蛋白结合蛋白-2mRNA的表达升高.经荧光素酶活性检测,得出紫杉醇诱导硫氧还蛋白结合蛋白-2基因启动子活性的增高.p38丝裂原激活蛋白激酶(p38 MAPK)抑制剂,SB203580抑制了紫杉醇诱导的硫氧还蛋白结合蛋白-2mRNA的表达.因此,紫杉醇抑制MCF-7细胞生长,并通过p38丝裂原激活蛋白激酶途径诱导硫氧还蛋白结合蛋白-2表达. The expression of paclitaxel inducing thioredoxin binding protein - 2 ( TBP - 2 ) and the inducing pathway of TBP- 2 in MCF- 7 cell are firstly discussed in this paper. MCF- 7 cells are treated with paclitaxel at different concentrations and the growth of MCF cells is inhibited. The expression of TBP - 2 induced by paclitaxel is examined by Real -PCR. The activity of TBP -2 reporter is increased by paclitaxel by the Luc assay. The expression of TBP -2 induced by paclitaxel is inhibited by p38 MAPKinase inhibitor, SB203580. Thus, paclitaxel inhibits MCF- 7 cell growth and induces TBP- 2 expression through the p38 MAPkinase pathway.
