摘要
应用胆固醇生物合成的体外酶反应筛选系统,从1000多株真菌中筛选出具有抑制胆固醇生物合成活性的SIPI-8917菌株;对SIPI-8917菌株的代谢产物进一步研究,应用丙酮浸泡、溶媒萃取、硅胶柱分离和LH20凝胶层析等方法,从其菌丝体中分离出SIPI-8917-Ⅰ化合物。通过元素分析和质谱,确定其分子式为:C22H20O7(分子量396.2905),综合UV、IR、1H-NMR、13C-NMR及HMBC和HMQC等图谱数据的解析,确定其结构为蒽环类化合物,与文献报道的1,3-二甲醚奥佛尼红素(1,3-Di-O-methylaverufin)结构一致。
Using a method for screening of cholesterol biosynthetic enzyme inhinitors from microorganisms in an enzyme system in vitro, a positive strain named SIPI8917 was obtained from more than 1000 fungi collected in our laboratory, whose metabolites exhibited inhibitory activities on cholesterol biosynthetic enzyme.Physiologically active compound SIPI8917Ⅰ was isolated from the mycelia of fungi SIPI8917 by acetone soaking, solvent extraction, silica gel column, LH20 column chromatography etc. Based on EIMS, FAB and HRMS, its formula was determined to be C_22H_20O_7 (MW 396.2905). By analyses of UV, IR, 1HNMR, 13 CNMR, HMQC and HMBC etc. spectra data, the structure of SIPI8917Ⅰ was elucidated and was identical with that of 1, 3-di-O-methylaverufin.
