RP-HPLC法测定制首乌和人参首乌胶囊中大黄素的含量

Determination of Emodin in Radix Polygoni Multiflori Preparata and Renshen Shouwu Capsules by RP-HPLC

目的：建立反相高效液相色谱法测定制首乌和人参首乌胶囊中大黄素的含量。方法：采用SPHERI-SORB ODS2（150mm×4.6mm,5μm）色谱柱,流动相为甲醇-0.1%磷酸（85∶15）,检测波长为254nm,柱温28℃,流速1.0mL.min-1。结果：大黄素进样量在0.03-0.20μg范围内与峰面积线性关系良好（r=0.9996）,平均回收率制首乌药材为97.5%,RSD为2.6%;人参首乌胶囊为98.0%,RSD=2.3%。结论：方法简便快捷,准确灵敏,重复性好,可用于原药材制首乌及其制剂人参首乌胶囊的质量控制。

Objective：To establish an RP-HPLC method for determination of the contents of emodin in Radix Polygoni multiflori Preparata and Renshen shouwu capsules.Methods：A SPHERISORB ODS2 column（4.6mm×150mm,5μm） was selected as analytical column.The mobile phase consisted of methanol-0.1% phosphoric acid（85 ∶ 15）.The temperature of column was 28℃.The flow rate was 1.0 mL·min-1.The detection wavelength was set at 254nm.Results：The calibration curve was linear with in a range of 0.03-0.20μg（r=0.9996）,The average recovery of Radix Polygoni multiflori Preparata and Renshenshouwu capsules were 97.5% with a RSD = 2.6% and 98.0% with a RSD = 2.3%. Conclution ：The method is simple ,rapid, accurate, sensitive and reproducible. It can be used for quality control of Radix Polygoni multiflofi Preparata and Renshen Shouwu capsules.
