Establishment of PCR-RFLP: a simple and rapidtechnique for HLA class Ⅱ DNA typing

目的建立一种简便、快速、价廉的ＨＬＡ－Ⅱ类基因分型方法，即ＰＣＲ－ＲＦＬＰ（ＲｅｓｔｒｉｃｔｉｏｎＦｒａｇｍｅｎｔＬｅｎｇｔｈＰｏｌｙｍｏｒｐｈｉｓｍ）。方法通过对４０例正常献血员及３０例系统性红斑狼疮（ＳＬＥ）患者ＨＬＡ－ＤＱＡ１基因的分型，介绍ＰＣＲ－ＲＦＬＰ的原理，具体方法及应注意的问题。结果正常献血员与ＳＬＥ患者有明显不同的基因型别。结论ＰＣＲ－ＲＦＬＰ分型方法更易为一般实验室所接受。

Objective\ To establish a simple, rapid and low-price genotyping method for HLAⅡ gene ie PCR-RFLP (Restriction Fragment Length Polymorphism). Methods\ The principle, method and warning of PCR-RFLP technique were presented by typing HLA-DQA1 gene of 40 donors and 30 patients with systemic lupus erythematosus. Results\ There was obvious difference gene typing between donors and patients with SLE. Conclusion\ The technique can be easily accepted by most lab compared to other genotyping methods.
