自然杀伤细胞的独立刺激信号——CD45分子交联

摘要 目的:探讨表面分子在自然杀伤(NK)细胞活化中的信号分子作用及其机制。方法:采用多种粘附分子单抗及配体刺激NK细胞,ELISA法定量检测刺激后NK细胞释放的γ干扰素(IFN-γ)作为NK细胞活化指标,观察各种粘附分子在NK细胞释放IFN-γ中的信号作用。结果:单一CD45单抗刺激即可使NK细胞活化;两种分别为IgG1,IgG2a亚类的CD45单抗及IgG2a亚类CD45单抗的F(ab)2均体现对NK细胞的独立刺激作用。结论:CD45分子介导的NK细胞活化与CD16-Fc段间相互作用无关。 Objective: To determine the identity of molecules and mechanism that mediate signal transduction and NK cell activation. Methods: We quantitated IFNγ concentration produced by NK cells that were stimulated with a panel of purified ligands and monoclonal antibodies specific for adhesion molecules. Results: AntiCD45 mAbs could specifically and independently stimulated IFNγ production in NK cells. The ability of CD45 mAbs to stimulate NK cells appears not to be dependent on CD16Fc partition interaction, as CD45 mAbs of both IgGl and IgG2a isotypes,both whole antibodies and its F(ab)2,were equally stimulatory. Conclusion: These observations suggest that CD45 crosslinking is the independent and unique stimuli for NK cell activation.
