摘要
目的:了解肝细胞生长因子(HGF)及其受体基因在人肝癌和癌旁肝组织中表达的变化规律,以及这种变化与肿瘤生物学行为的关系。方法:采用地高辛标记的HGF及其受体DNA探针,检测26例人原发性肝癌及其癌旁肝组织中HGF及其受体基因的表达。结果:HGF在人肝癌和癌旁肝组织中虽都有表达,但阳性率不及其受体基因表达,HGF受体在肝癌和癌旁肝组织中均呈过度表达,且肝癌组织中杂交信号明显高于癌旁肝组织,有肝内外转移者其表达更高。结论:HGF受体基因过度表达与肝癌的转移密切相关,HGF及其受体系统对调节肿瘤生长。
Objective: To study the changes of gene expression of hepatocyte growth factor (HGF) and hepatocyte growth factor receptor (HGFr) in hepatocellular carcinoma(HCC) tissue and nontumourous liver tissue and the relationship between these changes and the biological behavior of the tumor. Methods: The gene expression of HGF and HGFr in 26 cases of HCC tissue and nontumourous liver tissue was determined with digoxigeninlabled DNA probes. Results: The positive expression of HGF in HCC tissue was similar to that in their adjacent nontumourous liver tissue, but the positive rate of HGF expression was lower than HGFr gene expression. However, in the cases of metastasis, HGFr expression were higher. We found overexpression of HGFr in HCC tissue and their adjacent nontumourous liver tissue. Conclusion: There is closely relationship between overexpression of HGFr gene and metastasis of tumor, and the HGF and HGFr system plays an important role in regulating tumor growth and metastasis.
