摘要 目的:利用穿梭质粒pAd track CMV,探讨抑癌基因IL-24对宫颈癌细胞株Siha细胞生长和凋亡的影响。方法:将携带目的基因IL-24的穿梭质粒pAd track CMV转染入体外培养的宫颈癌细胞株Siha细胞,用MTT法检测细胞的生长增殖,用流式细胞术测定细胞周期及凋亡率。结果:经MTT法测定,3组Siha细胞OD值的总体差异有统计学意义(P<0.001),两两比较,IL-24组Siha细胞OD值明显低于空质粒组和阴性对照组(P<0.001),空质粒组和阴性对照组Siha细胞OD值则无明显差异(P=0.100);经流式细胞术测定,3组Siha细胞早期凋亡率的总体差异有统计学意义(P<0.05),两两比较,IL-24组Siha细胞早期凋亡率明显高于空质粒组和阴性对照组(P<0.05),空质粒组和阴性对照组Siha细胞的早期凋亡率无明显差异(P>0.05);与空质粒组和阴性对照组Siha细胞相比,IL-24组Siha细胞G0/G1期细胞增多(P<0.05),S期细胞减少(P<0.05),G2/M期细胞减少(P<0.05),空质粒组和阴性对照组则无明显差异(P>0.05)。结论:IL-24对宫颈癌细胞有抑制生长、诱导凋亡的作用。 Objective:To study the effect of tumor suppressor gene IL-24 on the cell growth and apoptosis of cervical cancer Siha cell line by using the shuttle vector pAd track CMV. Methods:The shuttle vector pAd track CMV that carried the objective gene of IL-24 was transfected into cervical cancer Siha cell line. Methyl thiazolyl tetrazolium (MTT) rapid photocolorimetric assay was used to examine the cell proliferation of Siha. Flow cytometry was used to examine the cell cycle and rate of apoptosis of Siha. Results : The OD values showed significant difference among the three groups of cells by MTT assay (P〈0.001). Compare to the other two groups,the OD values of IL-24 group was obviously reduced(P〈0. 001 ). There was no significant difference between the empty-vector group and the negative control group(P=0. 100). The apoptosis rates showed significant difference among the three groups of cells by flow cytometry (P〈0. 05). Compare to the other two groups, the apoptosis rate of IL-24 group was obviously reduced (P〈0.05),but no significant difference was shown between the empty-vector group and the negative control group (P〉0.05). In addition, compare to the other two groups, the number of cells in G0/G1 period was increased,the number of cells in S period was reduced (P 〈0.05 ), and the number of cells in G2/M period was reduced (P〈0.05). There was no significant difference between the empty-vector group and the negative control group (P〉0. 05). Conclusion : IL-24 gene can inhibit the growth and induce the apoptosis of cervical cancer Siha cell line.
